JCB • VOLUME 167 • NUMBER 2 • 2004 190 A chaperone feels the heat chaperone is supposed to keep its cool when temperatures get hot. But Syed Rizvi, Laura Mancino, Malini Raghavan (University of Michigan, Ann Arbor, MI), and colleagues show that calreticulin—a glycoprotein chaperone—starts to melt in the heat. The resulting structural changes actually improve its chaperone activity and may also occur transiently under normal conditions. Calreticulin is a stress-induced chaperone that helps glycoproteins such as MHC Class I molecules fold properly in the ER by binding to the target proteins’ sugar groups. The new results show that at high temperatures or low calcium levels, calreticulin can also bind independently of sugars and thus more effectively inhibit protein aggregation. Protein binding was accompanied by structural changes in calreticulin, including oligomerization at high temperatures. “It has remarkable conformational lability for a chaperone,” says Raghavan. “Its stability is like [that of] the substrates themselves.” The COOHterminal tail was found to be necessary to inhibit the oligomerized form, but its effect may be overcome at high temperature or low calcium so that the protein can help out when the ER is overwhelmed.

Numerical investigation of upward air-water annular, slug and bubbly flow regimes

In this paper, numerical investigation of upward two phase flow of air-water has been studied. Different conditions of flow regimes including annular, wispy annular, slug, churn and bubbly are simulated based on Hewitt and Roberts map, and a good agreement between the experimental data of the map and the numerical simulation has been observed. Accordingly, a proper CFD model in CFD software of ...

Two-Dimensional Numerical Simulations of Ultrasound in Liquids with Gas Bubble Agglomerates: Examples of Bubbly-Liquid-Type Acoustic Metamaterials (BLAMMs)

This work deals with a theoretical analysis about the possibility of using linear and nonlinear acoustic properties to modify ultrasound by adding gas bubbles of determined sizes in a liquid. We use a two-dimensional numerical model to evaluate the effect that one and several monodisperse bubble populations confined in restricted areas of a liquid have on ultrasound by calculating their nonline...

Direct Numerical Simulation of Bubbly Flows and Application to Cavitation Mitigation

The direct numerical simulation (DNS) method has been used to the study of the linear and shock wave propagation in bubbly fluids and the estimation of the efficiency of the cavitation mitigation in the container of the Spallation Neutron Source liquid mercury target. The DNS method for bubbly flows is based on the front tracking technique developed for free surface flows. Our front tracking hy...

Direct and Homogeneous Numerical Approaches to Multiphase Flows and Applications

We have studied two approaches to the modeling of bubbly and cavitating fluids. The first approach is based on the direct numerical simulation of gas bubbles using the interface tracking technique. The second one uses a homogeneous description of bubbly fluid properties. Two techniques are complementary and can be applied to resolve different spatial scales in simulations. Numerical simulations...

Surfactant effect on the multiscale structure of bubbly flows

Small amounts of surfactant can drastically change bubble behavior. For example, a bubble in aqueous surfactant solution rises much slower than one in purified water. This phenomenon is explained by the so-called Marangoni effect caused by a nonuniform concentration distribution of surfactant along the bubble surface. In other words, a tangential shear stress appears on the bubble surface due t...

Effective Medium Theory for Acoustic Waves in Bubbly Fluids near Minnaert Resonant Frequency

We derive an effective medium theory for acoustic wave propagation in bubbly fluid near Minnaert resonant frequency. We start with a multiple scattering formulation of the scattering problem of an incident wave by a large number of identical small bubbles in a homogeneous fluid. Under certain conditions on the configuration of the bubbles, we justify the point interaction approximation and esta...
